Biological Activity

Apabetalone (RVX-208) is a potent BET bromodomain inhibitor selective for BD2 with approximately 170-fold selectivity over BD1. By binding to BET bromodomains, apabetalone displaces BET proteins from chromatin and downregulates the transcription of pro-inflammatory genes and vascular adhesion molecules. This selective target inhibition increases apolipoprotein A-I expression and attenuates atherosclerosis-associated pathway signaling in vascular endothelial cells. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
