RS-127445 is a potent, selective serotonin 5-HT2B receptor antagonist with high affinity (pKi 9.5, pIC50 10.4) and over 1,000-fold selectivity over other serotonin receptors. By inhibiting 5-HT2B receptor signaling, RS-127445 blocks 5-HT-evoked inositol phosphate formation and intracellular calcium mobilization. Downstream, this target inhibition effectively attenuates 5-HT-induced smooth muscle contraction and alters gastrointestinal motility phenotypes. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
