Biological Activity

RP-6306 (lunresertib) is an orally bioavailable, selective inhibitor of protein kinase, membrane-associated tyrosine/threonine 1 (PKMYT1). By suppressing PKMYT1 kinase activity, RP-6306 induces unscheduled activation of CDK1 selectively in CCNE1-overexpressing cancer cells. This premature CDK1 activation forces cells into early mitosis during DNA synthesis, leading to profound DNA damage, replication stress, and synthetic lethal tumor cell death. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
