Rolapitant (SCH-619734) is a selective, competitive antagonist of the human neurokinin-1 (NK1) receptor with a Ki of 0.66 nM. It exhibits greater than 1000-fold selectivity for the human NK1 receptor over human NK2 and NK3 subtypes. By blocking substance P binding to NK1 receptors, rolapitant suppresses neurokinin signaling, thereby inhibiting central emetic pathways and demonstrating potent antiemetic activity in preclinical models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
