Rofecoxib (MK-0966) is a potent and selective cyclooxygenase-2 (COX-2) inhibitor with an IC50 of 18 nM. By selectively inhibiting COX-2 activity, it blocks the conversion of arachidonic acid to prostaglandin E2 (PGE2) without significantly altering COX-1 function. This suppression of prostaglandin synthesis suppresses downstream inflammatory signaling pathways, leading to reduced cellular inflammation and pain signaling responses in preclinical cell and animal models. - Preheat media/saline to 37°C.
