Ro 67-7476 is a potent positive allosteric modulator (PAM) of metabotropic glutamate receptor 1 (mGluR1), enhancing glutamate-evoked responses in calcium mobilization assays with an EC50 of. It also exhibits intrinsic agonist activity by inducing ERK1/2 phosphorylation with an EC50 of. Potentiation of mGluR1 signaling modulates intracellular calcium dynamics and downstream MAPK/ERK pathways, regulating neuronal excitability and synaptic function. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
