RMC-5127 is an orally bioavailable, mutant-selective tri-complex inhibitor targeting the GTP-bound (ON) state of RAS G12V. By non-covalently binding cyclophilin A, it forms a high-affinity tri-complex with active RAS G12V, sterically blocking effector recruitment and suppressing downstream RAS signaling pathways. Consequently, RMC-5127 demonstrates potent anti-tumor activity in preclinical KRAS G12V-mutant non-small cell lung, pancreatic, and colorectal cancer xenograft models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
