Rimiducid (AP1903) is a synthetic chemical dimerizer that selectively binds mutant FKBP12(F36V) fusion proteins with subnanomolar affinity while showing minimal binding to endogenous wild-type FKBP12. By inducing the homodimerization or oligomerization of FKBP-containing chimeric receptors or enzymes, such as Fas or caspase signaling constructs, rimiducid activates downstream pro-apoptotic pathways. This targeted dimerization triggers potent, dose-dependent apoptosis in cells expressing dimerizer-dependent constructs, serving as an inducible cell safety switch. [1]
