Biological Activity

RGFP966 is a selective histone deacetylase 3 (HDAC3) inhibitor with an IC50 of 0.08 μM in cell-free assays, displaying >200-fold selectivity over other HDAC isoforms. By specifically inhibiting HDAC3, RGFP966 enhances histone acetylation and modulates downstream gene transcription pathways. This leads to promoted synaptic plasticity, improved memory consolidation, and anti-inflammatory cellular responses in various preclinical models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
