Resibufogenin is a bioactive bufadienolide component of huachansu with potent anticancer activity. It triggers necroptosis by upregulating receptor-interacting protein kinase 3 (RIP3) and promoting the phosphorylation of mixed lineage kinase domain-like protein (MLKL) at Ser358. Furthermore, resibufogenin induces reactive oxygen species (ROS) accumulation and activates caspase-3 and caspase-8 to promote apoptosis, while downregulating PI3K, p-AKT, p-GSK3β, cyclin D1, and β-catenin signaling pathways. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
