Biological Activity

GS-5734 (Remdesivir) is a monophosphoramidate prodrug of an adenosine analog that acts as a broad-spectrum antiviral inhibitor targeting viral RNA-dependent RNA polymerase. Upon intracellular metabolization to its active nucleoside triphosphate derivative, it competes with natural ATP for incorporation into nascent viral RNA transcripts, leading to delayed chain termination and inhibition of viral RNA synthesis. Consequently, GS-5734 suppresses replication across diverse RNA viruses, including Ebola virus and coronaviruses. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
