(R)-baclofen is a selective agonist of the metabotropic GABAB receptor that functions as the active enantiomer of baclofen. Activation of GABAB receptor complexes inhibits adenylyl cyclase activity and downregulates voltage-gated calcium channels while activating inwardly rectifying potassium channels. These signaling cascades depress presynaptic neurotransmitter release and hyperpolarize postsynaptic neuronal membranes, leading to suppressed excitatory synaptic transmission and attenuated muscle spasticity.
