Gedatolisib (PKI-587) is a potent dual inhibitor of PI3Kα, PI3Kγ, and mTOR. It suppresses the PI3K/AKT/mTOR signaling pathway by inhibiting AKT phosphorylation at T308 and S473. This inhibition leads to PARP cleavage, induction of apoptosis, and suppression of cellular proliferation. Gedatolisib exhibits potent antitumor activity in various human cancer xenograft models, including breast, colon, lung, and glioma tumors. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
