PKI-402 is a potent dual pan-PI3K/mTOR inhibitor targeting PI3Kα (IC50 2 nM), PI3Kβ, PI3Kγ, PI3Kδ, and mTOR, with high activity against PI3Kα mutants E545K and H1047R. It suppresses downstream AKT/mTOR signaling by blocking AKT phosphorylation at T308, triggering apoptosis and driving tumor cell growth inhibition and xenograft tumor regression in vivo. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
