Pitstop 2 is a cell-permeable inhibitor targeting the terminal domain of clathrin, disrupting its interaction with amphiphysin. By binding to the clathrin terminal domain, it inhibits both clathrin-dependent endocytosis and clathrin-independent endocytosis. Disruption of endocytosis impairs receptor internalization, synaptic vesicle recycling, and intracellular membrane trafficking, thereby modulating cellular nutrient uptake and signaling pathways. [1]
