Biological Activity

Pitolisant hydrochloride is a potent, high-affinity competitive antagonist of the human histamine H3 receptor (Ki = 0.16 nM) and acts as an inverse agonist (EC50 = 1.5 nM). By blocking presynaptic H3 autoreceptors, it inhibits autoinhibition of histamine neurons and enhances histaminergic signaling in the central nervous system. This activation increases cortical histamine release and modulates downstream neurotransmitter pathways involved in promoting wakefulness in preclinical models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
