Biological Activity
PF-562271 is an ATP-competitive, reversible inhibitor of focal adhesion kinase (FAK, IC50 1.5 nM) in cell-free assays. It demonstrates approximately 10-fold selectivity for FAK over Pyk2 and broad selectivity against other protein kinases. By inhibiting FAK autophosphorylation and disrupting downstream integrin signaling, PF-562271 suppresses cancer cell migration, invasion, proliferation, and tumor growth in multiple preclinical models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
