PF-07220060 (CDK4/6-IN-6) is a selective cyclin-dependent kinase (CDK) inhibitor targeting CDK4/Cyclin D1 over CDK6/Cyclin D3. By selectively targeting CDK4 within the cyclin D–CDK4 pathway, it inhibits retinoblastoma protein phosphorylation to induce cell cycle arrest in the G1 phase. This mechanism blocks downstream E2F-mediated gene transcription, leading to potent suppression of tumor cell proliferation. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
