Biological Activity

PF-06446846 hydrochloride is an orally active, selective small-molecule inhibitor of proprotein convertase subtilisin/kexin type 9 (PCSK9) translation with an IC50 of 0.3 µM in Huh7 cells. It engages the human 80S ribosome to stall translation around codon 34 of the nascent chain. This selective translational arrest blocks PCSK9 synthesis, thereby reducing circulating PCSK9 levels and lowering plasma total cholesterol in vivo. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
