Biological Activity

PF-04957325 is a selective phosphodiesterase-8 inhibitor targeting PDE8A (IC50 0.7 nM) and PDE8B (IC50 0.3 nM). By inhibiting PDE8-mediated cAMP hydrolysis, the compound elevates intracellular cAMP levels and significantly potentiates steroidogenesis in adrenal cells. In cellular immune responses, PF-04957325 modulates the PDE8-controlled cAMP/PKA signaling pathway to suppress the adhesion, locomotion, and endothelial interaction of inflammatory CD4+ T effector cells.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
