Biological Activity
PF-04691502 is an ATP-competitive dual PI3K and mTOR inhibitor targeting PI3Kα, PI3Kβ, PI3Kδ, PI3Kγ, and mTOR. By blocking the PI3K/AKT/mTOR signaling pathway, it suppresses phosphorylation of downstream effectors including AKT, p70S6K, and 4EBP1. This inhibition induces G1 cell cycle arrest, promotes p27 Kip1 expression, and triggers apoptosis in cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
