ONX-0914 (PR-957) is a selective immunoproteasome inhibitor targeting the immunoproteasome subunit LMP7 (β5i) with minimal cross-reactivity for the constitutive proteasome. By selectively inhibiting immunoproteasome catalytic activity, ONX-0914 blocks the production of proinflammatory cytokines such as IL-23, IL-17, and TNF-α. This inhibition modulates immune signaling pathways, suppresses inflammatory cell activation, and attenuates disease progression in experimental models of autoimmune inflammation and arthritis. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
