Biological Activity

NU7441 is a potent and selective DNA-dependent protein kinase (DNA-PK) inhibitor (IC50 = that also inhibits mTOR (IC50 = and PI3K (IC50 =. By suppressing DNA-PK-mediated non-homologous end joining repair of double-strand breaks, it disrupts DNA damage repair pathways, promoting G2/M cell cycle arrest and potentiating cytotoxicity induced by ionizing radiation and topoisomerase II inhibitors. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
