NSC232003 is a potent, cell-permeable UHRF1 inhibitor that specifically binds to the 5-methylcytosine (5mC) binding pocket within the SRA domain of UHRF1. Disruption of UHRF1 functionality blocks the critical DNMT1/UHRF1 protein-protein interaction necessary for DNA methylation maintenance during cell replication. Consequently, treatment with NSC232003 suppresses DNA methylation inheritance and induces global DNA cytosine demethylation in cancer cellular models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
