NSC 23766 trihydrochloride is a selective inhibitor of Rac GTPase, targeting Rac activation mediated by guanine nucleotide exchange factors (GEFs) with an IC50 of ~in cell-free assays, without inhibiting Cdc42 or RhoA. By preventing GEF binding and Rac activation, NSC 23766 suppresses downstream Rac signaling pathways, leading to the inhibition of cell proliferation, migration, invasion, and cell cycle progression in various cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
