Biological Activity

NLRP3/AIM2-IN-3 is a potent small-molecule inhibitor targeting NLRP3 and AIM2 inflammasome-mediated pyroptosis. In THP-1 macrophages stimulated with LPS and nigericin, it effectively suppresses pyroptosis with species-specific activity. Mechanistically, the compound disrupts the molecular interaction between NLRP3 or AIM2 and the adaptor protein ASC, thereby preventing ASC oligomerization and blocking downstream inflammasome assembly. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
