Niclosamide ethanolamine is an anthelmintic agent, mild mitochondrial uncoupler, and potent inhibitor of the STAT3 signaling pathway (IC50 =. It suppresses PI3K-mTORC1 signaling to block fibroblast proliferation and inhibits TGF-β1-induced epithelial-mesenchymal transition and extracellular matrix accumulation via the mTORC1-4EBP1 axis. Furthermore, it activates noncanonical autophagy to sensitize fibroblasts to apoptosis, conferring renoprotection in diabetic and nondiabetic kidney disease models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
