Biological Activity

Nepicastat HCl is a potent and selective dopamine β-hydroxylase inhibitor that targets bovine dopamine β-hydroxylase (IC50 8.5 nM) and human dopamine β-hydroxylase (IC50 9 nM). By inhibiting dopamine β-hydroxylase, it blocks the enzymatic conversion of dopamine to norepinephrine. This inhibition alters catecholamine pathways, leading to dose-dependent depletion of tissue noradrenaline content accompanied by increased dopamine levels and elevated dopamine-to-noradrenaline ratios.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
