Biological Activity

Necrosulfonamide is a potent inhibitor of mixed lineage kinase domain-like protein (MLKL) that specifically blocks necrosis. By directly binding to MLKL and preventing its oligomerization and plasma membrane translocation, Necrosulfonamide suppresses RIP3-dependent necroptotic cell death downstream of TNF-alpha signaling. This compound effectively inhibits necroptosis in human cell models without interfering with apoptosis pathways, serving as a critical tool for dissecting necroptotic signaling mechanisms. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
