Biological Activity

Firsocostat (ND-630, GS-0976) is an orally bioavailable, reversible acetyl-CoA carboxylase inhibitor targeting human ACC1 and human ACC2. By binding the biotin carboxylase domain of ACC, firsocostat blocks the conversion of acetyl-CoA to malonyl-CoA, effectively inhibiting de novo lipogenesis. Consequently, this target inhibition reduces hepatic steatosis, decreases fatty acid synthesis, and improves insulin sensitivity in liver tissue models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
