JPH203 (Nanvuranlat) is a selective L-type amino acid transporter 1 (LAT1) inhibitor that suppresses leucine uptake with an IC50 of 0.06 µM. By blocking LAT1-mediated amino acid transport, JPH203 inhibits HT-29 cell proliferation with an IC50 of 4.1 µM. Disruption of intracellular leucine supply leads to the downregulation of mTORC1 signaling, ultimately inducing growth arrest and apoptosis in colon and oral cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
