ML347 is a selective bone morphogenetic protein (BMP) receptor inhibitor that targets ALK2 (IC50 of and ALK1 (IC50 of, displaying over 300-fold selectivity for ALK2 over ALK3. By inhibiting ALK1 and ALK2 kinase activity, ML347 blocks downstream Smad1/5/8 phosphorylation within the BMP signaling pathway. This blockade modulates cellular phenotypes such as osteogenic differentiation and BMP-dependent gene expression. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
