ML324 is a selective inhibitor of jumonji histone demethylase (JMJD2) with an IC50 of. By blocking KDM4/JMJD2 demethylase activity, ML324 prevents the demethylation of methylated histone substrates, promoting hypermethylation of H3K9 and H3K36. This elevation in histone methylation suppresses downstream oncogenic transcriptional programs, leading to impaired cell proliferation in prostate cancer cells and inhibition of viral gene expression and infection.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
