Niraparib tosylate is a selective poly(ADP-ribose) polymerase inhibitor targeting PARP1 (IC50 3.8 nM) and PARP2 (IC50 2.1 nM). By suppressing PARP enzymatic activity, it enhances the formation of persistent PARP-DNA complexes and blocks base excision repair. This accumulation of unrepaired single-strand breaks leads to double-strand DNA damage during replication, ultimately inducing severe genomic instability, apoptosis, and cell death in homologous recombination-deficient cells.
