Mizolastine is a potent histamine H1-receptor antagonist with an IC50 of 47 nM. By targeting histamine H1 receptors, it effectively blocks downstream H1 receptor-mediated signaling pathways, inhibiting histamine-induced vascular permeability, smooth muscle contraction, and the release of pro-inflammatory mediators. Consequently, mizolastine suppresses allergic airway responses, cutaneous wheal-and-flare reactions, and allergic phenotype manifestations associated with seasonal allergic rhinitis and hives.
