MG-101 (ALLN) is a cell-permeable peptide aldehyde that acts as a potent inhibitor of cysteine proteases, including calpains and cathepsins, as well as the 20S proteasome. By inhibiting proteasomal and lysosomal protein degradation pathways, MG-101 prevents the cleavage of key cellular substrates, leading to the accumulation of regulatory proteins such as cyclins and IκBα. Consequently, treatment with MG-101 suppresses cell cycle progression, induces apoptosis, and modulates downstream cell signaling events. [1]
