Mevastatin (ML-236B) is a competitive inhibitor of HMG-CoA reductase, displaying a binding affinity approximately 10,000 times higher than the endogenous substrate HMG-CoA. By inhibiting HMG-CoA reductase, it blocks the rate-limiting step of the mevalonate pathway, suppressing cholesterol biosynthesis and depleting downstream isoprenoid intermediates. This pathway inhibition leads to decreased cell proliferation, cell cycle arrest, and induction of apoptosis in various cellular models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
