Menthol is a natural analgesic compound and TRPM8 receptor agonist that modulates calcium ion influx in neuronal membranes. By activating the cold- and menthol-sensitive TRPM8 channel in sensory neurons, menthol stimulates cold receptor signalling while simultaneously suppressing voltage-gated calcium currents. Downstream activation of this pathway leads to thermosensation modulation, local hypalgesia, and anti-nociceptive phenotypes in neuronal tissues. [1][2]
