MBoIN179 is a small-molecule modulator that selectively disrupts the transmembrane-domain interaction between the anti-apoptotic protein MCL1 and the pro-apoptotic protein BOK. By releasing BOK from MCL1-mediated sequestration, MBoIN179 promotes BOK-dependent oligomerization and mitochondrial outer-membrane permeabilization. This disinhibition of BOK activates downstream apoptotic signaling pathways, leading to mitochondrial dysfunction and the induction of apoptotic cell death. [1]
