Macitentan is an orally active dual endothelin receptor antagonist targeting ETA (IC50 = and ETB (IC50 =. By blocking endothelin-1 binding to its receptors, macitentan inhibits receptor-mediated intracellular calcium mobilization, vascular smooth muscle cell contraction, and cell proliferation. In preclinical animal models, macitentan effectively attenuates pulmonary arterial hypertension and reduces vascular remodeling and arterial wall hypertrophy. [1]
