Clinical Trials

Multiple clinical trials across Phase 2, Phase 3, and Phase 4 have evaluated lumiracoxib for managing pain, osteoarthritis, rheumatoid arthritis, acute gouty arthritis, and arthralgia, alongside pharmacodynamic assessments in healthy volunteers and patients with controlled hypertension or chronic kidney failure. Sponsored primarily by pharmaceutical industry leaders such as Novartis and medical institutions like Hospital Central Sur de Pemex, the majority of these trials are completed, though a subset was terminated.

Compliance for Clinical Use

Mechanism and Biochemical Profile

Lumiracoxib is a highly selective cyclooxygenase-2 inhibitor (Ki = 0.06 μM) that preferentially targets COX-2 over COX-1 (Ki = 3 μM), thereby blocking the enzymatic conversion of arachidonic acid into pro-inflammatory prostaglandins. By suppressing downstream prostaglandin E2 synthesis, Lumiracoxib reduces localized inflammatory signaling and nociceptor sensitization, providing therapeutic alleviation of joint pain and tissue inflammation in clinical conditions such as osteoarthritis and rheumatoid arthritis.

Appendix RUO and cGMP Quality Standards

Quality Dimension RUO (Research Use Only) cGMP (Current Good Manufacturing Practice)
Clinical Applicability Prohibited in human clinical trials or medical diagnostics. Mandatory for human clinical trials (Phase I–III) and therapies.
Regulatory Status Non-regulated grade; exempt from drug manufacturing laws. Legally enforced by health authorities (e.g., FDA, EMA, NMPA).
Facility Environment Unclassified analytical or research laboratories. Validated Cleanrooms (ISO Class 5–8) with continuous monitoring.
Quality Control Basic purity and activity testing. Rigorous safety release testing (Sterility, Endotoxin, Mycoplasma).
Process Validation Basic equipment calibration; no process validation required. Full qualification (IQ/OQ/PQ) and complete batch records.
Quality Assurance Vendor self-declared without required formal QMS. Mandatory QA/QC unit, Change Control, CAPA, and vendor audits.
Regulatory Impact High risk of IND rejection if used as a critical raw material. Required for IND/NDA filings, supported by Drug Master Files (DMF).
