Lumefantrine (benflumetol) is a synthetic antimalarial agent that acts as an inhibitor of heme polymerization in Plasmodium parasites. By binding to hemin, it blocks the conversion of toxic free heme into non-toxic β-hematin within the parasite digestive vacuole. This accumulation of free heme disrupts membrane structure and function, inhibiting nucleic acid and protein synthesis to induce parasite cell death and suppress Plasmodium falciparum growth. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
