Leupeptin Hemisulfate is a reversible inhibitor of serine and cysteine proteases, selectively targeting cathepsin B (Ki 6 nM), calpain (Ki 10 nM), trypsin (Ki 35 nM), plasmin (Ki 3.4 μM), and kallikrein (Ki 19 μM). By blocking intracellular protein degradation and lysosomal proteolysis, it prevents proteasomal-independent protein turnover, modulates calcium-dependent signaling, and impairs autophagic degradation in various cellular models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
