Lenalidomide (CC-5013) is a TNF-α secretion inhibitor in PBMCs) and a ligand of the cereblon (CRBN) E3 ubiquitin ligase. Binding to the CRBN-CRL4 ubiquitin ligase complex induces the selective ubiquitination and proteasomal degradation of lymphoid transcription factors IKZF1 and IKZF3. Downstream, lenalidomide reduces VEGF expression, promotes cleaved caspase-3 expression, triggers G1 cell cycle arrest, and induces apoptosis in malignant cells. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
