LDC000067 is a highly selective inhibitor of cyclin-dependent kinase 9 (CDK9, IC50 44 nM). By inhibiting the CDK9 subunit within the positive transcription elongation factor b (P-TEFb) complex, LDC000067 blocks Ser2 phosphorylation of the RNA polymerase II C-terminal domain. This inhibition suppresses de novo gene transcription, downregulates short-lived anti-apoptotic proteins such as MCL-1, and significantly impairs cancer cell survival and proliferation.
