Sacubitril/valsartan (LCZ696) is an orally bioavailable, dual-acting angiotensin receptor-neprilysin inhibitor (ARNi) comprising the angiotensin II type 1 (AT1) receptor antagonist valsartan and the neprilysin inhibitor prodrug sacubitril in a 1:1 molar ratio. By simultaneously blocking AT1 receptor-mediated vasoconstriction and inhibiting neprilysin-mediated degradation of natriuretic peptides, it enhances cGMP signaling, promotes vasodilation, and suppresses cardiac remodeling and fluid retention.
