Biological Activity

Larazotide (AT-1001) is a synthetic octapeptide tight junction regulator and antagonist of zonulin. By blocking zonulin receptor binding, larazotide prevents tight junction disassembly and actin cytoskeleton rearrangement, thereby maintaining intestinal epithelial barrier integrity. This inhibition blocks the paracellular passage of luminal antigens such as gliadin peptides across the mucosal barrier, suppressing downstream antigen trafficking and inflammatory signaling cascades in preclinical models of intestinal hyperpermeability.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
