K02288 is a potent and selective type I bone morphogenetic protein (BMP) receptor inhibitor targeting ALK2, ALK1, and ALK6, with weaker activity against ALK3, ALK4, ALK5, and ActRIIA. By inhibiting BMP receptor kinase activity, K02288 blocks downstream Smad1/5/8 phosphorylation and suppresses BMP-induced transcriptional activity and target gene expression, modulating osteogenic differentiation and vascular signaling pathways. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
