K-604 dihydrochloride is a potent, selective inhibitor of human ACAT-1 (IC50 0.45 μM) over human ACAT-2 (IC50 102.85 μM), exhibiting 229-fold selectivity with a Ki of 0.378 μM. It suppresses macrophage cholesterol esterification (IC50 68.0 nM) and enhances cholesterol efflux. Consequently, K-604 blocks foam cell accumulation and attenuates fatty streak lesion formation in vivo without altering plasma cholesterol levels. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
