JW55 is a potent and selective canonical Wnt pathway inhibitor that functions by targeting the PARP domain of tankyrase 1 and tankyrase 2 (TNKS1/2). Inhibition of TNKS1/2 by JW55 stabilizes Axin proteins, promoting the degradation of cytosolic β-catenin and suppressing Wnt/β-catenin target gene expression. Downstream, this attenuation of canonical Wnt signaling suppresses cell proliferation and impairs colony formation in colorectal cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
